Identification of Campylobacter jejuni and Campylobacter coli from refrigerated chicken carcasses by Multiplex PCR

Campylobacter jejuni and C. coli are the most frequently identified thermophilic species in cases of human enteritis caused by the consumption of food products made with undercooked chicken or cross-contamination during handling of raw food products. The biochemical differentiation of C. jejuni and C. coli shows inconsistencies, such as hippurate hydrolysis and sensitivity to nalidixic acid and cephalothin. Consequently, the use of molecular methods is necessary to identify these species. The objective of this study was to identify C. jejuni and C. coli isolates from refrigerated chicken carcasses using multiplex PCR. For this purpose, the genes encoding a 160-bp subunit of oxidoreductase and the 894-bp ceuE virulence gene specific for C. jejuni and C. coli, respectively, were concurrently subjected to PCR. Of the 21 isolates assessed biochemically, 19 (90.48%) were identified as C. jejuni, one (4.76%) as C. coli, and one (4.76%) was not identified. Multiplex PCR confirmed the presence of 90.48% C. jejuni and 9.52% C. coli. The proposed approach was rapid, sensitive, and specific and is a good alternative for routine clinical testing and epidemiological studies.Downloads
